Tripeptide (copper peptide) is formed by three amino acids, and tripeptide is also called blue copper peptide; glycyl-L-histidyl-L-lysine. Tripeptide is a ternary molecule composed of three amino acids connected by two peptide bonds. It can effectively block the nerve conduction of an acetylcholine substance, relax muscles, and improve dynamic wrinkles. Peptides are composed of some amino acids with a specific sequence connected by amide bonds. Two amino acids are called dipeptides, three amino acids are called tripeptides, and so on. Even if the amino acids are exactly the same, if the arrangement and connection are different, they will form peptides with completely different structures. Tripeptide-copper, copper is a trace element required to maintain body functions (2 mg per day), with many and complex functions, and is an element required for the action of various cell enzymes. Because there are many important enzymes in the human body and on the skin that need Cu ions, these enzymes play a role in connective tissue formation, antioxidant defense, and cell respiration. Cu ions also play a signaling function and can affect cell behavior and metabolism. In terms of the role of skin tissue, it has the functions of anti-oxidation, promoting collagen proliferation, and assisting wound healing.
Basic properties
Copper peptides are unstable in an acidic environment, will release copper ions, and lose their function. Therefore, it is generally not used in acidic cosmetics or when used with acidic cosmetics, it needs to be used at an interval. It is easy to hydrolyze under oxidative conditions and can be stored for two weeks at 60°C in water or a buffer solution of pH 4.5-7.4. The distribution coefficient in octanol-phosphate buffered saline shows that GHK-Cu is highly hydrophilic and is not very stable under diacetyl phosphate conditions with negatively charged lipids.
Copper peptide powder must be stored in a cool, dark and clean place to ensure a shelf life of at least 12 months. For long-term storage, it is recommended to store at 4°C, and the shelf life can be extended to at least 24 months.
Application
1. In the form of skin care products:
CHK-Cu has multiple functions, including the role of messenger and the role of carrier and transport. Secondly, it has the advantages of being mild, non-irritating and safe, so it has high application value in topical skin care products.
(1) Anti-aging and anti-wrinkle effects: After copper peptide enters the skin, it releases signals to induce fibroblasts to become collagen and promote collagen production. At the same time, copper ions are carried into the cells and released after encountering enzymes. Copper ions are very important components in many enzymatic reactions. Researchers have done a lot of research on the anti-aging and anti-wrinkle effects of copper peptides. In vitro experiments have shown that its ability to stimulate collagen precursor synthesis is stronger than anti-aging ingredients such as retinoic acid and vitamin C. However, the anti-wrinkle effect is not as fast as vitamin A, and it takes several months of continuous use to feel the effect. However, compared with vitamin A, there is relatively little research data on copper peptides. Much of the available data comes from the inventor Picket and lacks independent third-party verification.
(2) Promote wound healing, repair damaged skin, and improve skin density and toughness: Copper peptides can promote the growth, division and differentiation of nerve cells and immune-related cells, effectively promote wound healing and hair growth, and repair damaged skin. Skin with minor trauma, damage, sensitive and fragile skin can be repaired by using products containing copper peptides.
(3) Antioxidant: The antioxidant effect of blue copper peptides has been confirmed in in vitro and animal experiments. It can block the formation of reactive oxygen species, remove various peroxides, protect keratinocytes from ultraviolet damage, and block free radical damage to the liver, thus protecting the liver. Blue copper peptide has excellent antioxidant effects, which can brighten the skin tone, refine the skin, and prevent skin aging.
(4) Usage and dosage: Skin repair peptide-1 is a copper peptide powder product, which can be added in the final stage of cosmetic production. The preparation temperature must be below 40°C. The recommended concentration is 500ppm-2,000ppm.
2. In the form of pharmaceutical preparations or dietary supplements
GHK-Cu microspheres coated with hydroxypropyl cellulose are used as colon-targeted release drugs. The release of GHK-Cu is affected by the degree of cross-linking and the drug loading. The microspheres release 50-80% of the drug within 4 hours. These studies suggest that GHK-Cu can be used as a dietary supplement.
